液相色谱-串联质谱法同时测定三七中508种农药残留

三七为五加科人参属植物三七Panaxnotoginseng(Burk)F.H.Chen的干燥根及根茎,主产于云南省文山州。其具有散瘀止血、消肿定痛之功效,是预防和治疗心脑血管疾病的首选药材,也是注射用血栓通、云南白药、片仔癀等药品的主要原料[1]。近年来三七的规模化人工种植日益普遍,在长达三年的生长过程中,三七易遭受各种病虫草害,因此种植过程中不可避免地使用农药。农药的过量或违规使用会导致三七药材中有农药残留,影响用药安全[2-3]。农药残留问题成为制约三七等药材走向国际市场的主要瓶颈之一。因此,亟需建立简便、高效的三七中农药残留的高通量检测方法,为农药的科学使用及政府监管提供技术支撑。

近年来,随着我国对中药材农药残留安全的重视,新的检测方法不断出现,如季申等[4-5]采用传统色谱方法分别测定了多种中药材的56种有机氯类农药与57种有机磷农药,分别采用电子捕获检测器与火焰光度检测器测定该两类农药,气相色谱双塔双柱法对阳性结果进行确证。但色谱方法需对农药进行色谱分离,单次进样可测定的农药数量仅几十种。近年来随着质谱技术发展,串联三重四极杆质谱在农药残留测定领域发挥着越来越重要的作用。较色谱方法而言,质谱法依据各农药的特征离子对定性定量,无需对各农药进行色谱分离,单次进样可测定的农药数量大大提高,且专属性好,针对中药材等复杂样本的抗基质干扰能力强。如季申等[6]建立了多种中药材中74种农药残留的液相色谱-串联质谱检测方法,黄莉莉等[7]建立了桑叶中100种农药残留的液相色谱-串联质谱测定方法,两种方法虽然测定农药种类较多,但前处理步骤较多,耗时费力,影响方法整体效能。

美国农业部Lehotay等[8]开发的QuEChERS前处理方法是对食品中农药残留测定技术的一次较大推进,该方法简便、快速、高效,依托该技术的食品中高通量多残留检测方法不断出现[9-14]。在中药材农残测定方面,季申等[15]采用气相色谱-串联三重四极杆质谱法实现了单次进样对黄芪中238种农药残留的检测,针对中药材特点对QuEChERS提取方法进行了改良。季申课题组还开发了中药中227种农药残留的气相色谱-串联三重四极杆质谱与液相色谱-串联三重四极杆质谱测定方法,采用缓冲盐QuEChERS方法提取,分散固相萃取净化,该方法已被中国药典2015年版四部通则[16]收载。然而中国药典所检测农药数量仍与食品安全国家标准有一定差距,如庞国芳等[17]针对桑枝、金银花、枸杞子和荷叶4种药材,开发了测定413种农药及相关化学品残留量的液相色谱-串联质谱法。为更好地满足中药材农药残留的监管需要,亟需进一步扩增检测农药数量。

本文在中国药典2015年版四部通则方法基础上,大幅扩增了农药数量,以改良的QuEChERS方法提取,分散固相萃取法(d-SPE)净化,采用液相色谱-串联三重四极杆质谱为检测手段,内标法定量,首次建立了单次进样可在20 min内测定三七中508种农药残留的高通量检测方法。

1 实验部分

1.1 仪器与试剂

Agilent 1290超高效液相色谱(美国Agilent公司),AB SCIEX 6500三重四极杆质谱(美国AB SCIEX公司);N-EVAPTM112型氮吹仪(美国Organomation公司);IKA KS260C振荡器、IKA MS 2涡旋混合器(德国IKA公司);UV2550紫外分光光度计(日本岛津公司);旋转蒸发仪(瑞士BÜCHI 公司)。

农药标准品均购于德国Dr.Ehrenstorfer GmbH公司;乙腈、甲醇均为色谱纯(美国Merck公司);醋酸(分析纯,国药集团上海化学试剂公司);甲酸、甲酸铵均为色谱纯(美国Sigma-Aldrich公司)。净化材料:N-丙基乙二胺(PSA)、石墨化碳黑(GCB)、十八烷基键合硅胶(C18)、硅胶(Silica)、提取管(50 mL,预装6 g无水硫酸镁、1.5 g无水醋酸钠)、净化管(15 mL,预装300 mg PSA、300 mg C18、90 mg GCB、600 mg Silica、900 mg无水硫酸镁)均购于天津博纳艾杰尔科技有限公司。

1.2 标准溶液的配制

精密称取各农药标准品及内标适量,用乙腈或甲苯分别制成1 000 μg/mL的储备溶液;精密吸取农药储备溶液适量,用含0.05%醋酸的乙腈稀释成100 μg/L和1 000 μg/L的混合标准溶液;精密量取内标储备溶液适量,用乙腈稀释成6 μg/mL的溶液。

称取三七空白样品6份,每份3 g,按照“1.4 ”方法进行提取、净化和离心后,取上清液5 mL,置氮吹仪上于40 ℃水浴浓缩至约0.4 mL,分别加入50、100 μL的100 μg/L混合标准溶液,以及50、100、200、400 μL的1 000 μg/L混合标准溶液,用乙腈定容至1 mL,涡旋混匀,过0.22 μm微孔滤膜,取续滤液,得质量浓度分别为5、10、50、100、200、400 μg/L的系列基质混合标准工作溶液。

1.3 分析条件

1.3.1 色谱条件色谱柱:Agilent Poroshell 120 EC-C18(3.0 mm×150 mm,2.7 μm);流动相:A为0.05%甲酸+5 mmol/L甲酸铵水溶液;B为0.05%甲酸+5 mmol/L甲酸铵甲醇。梯度洗脱程序:0~1.0 min,95%A;1.0~4.0 min,95%~40%A;4.0~8.0 min,40%~36%A;8.0~8.5 min,36%~32%A;8.5~9.0 min,32%~25%A;9.0~16.0 min,25%~5%A;16.0~20.0 min,5%A;20.0~20.1 min,5%~95%A。流速:0.40 mL/min;进样量:2.0 μL。

1.3.2 质谱条件离子源:电喷雾离子源(ESI);扫描方式:正离子模式;检测方式:多反应监测(MRM)模式;电喷雾电压:5 500 V;雾化气压力:344.8 kPa;辅助气压力:344.8 kPa;气帘气压力:137.9 kPa;碰撞气压力:48.3 kPa;离子源温度:500 ℃;扫描时间:10 ms;碰撞室入口电压:10 V;碰撞室出口电压:12 V;监测离子对及其质谱参数见表1。

1.4 样品前处理

供试品粉碎后过50目筛,取约3 g(精确至0.01 g)置50 mL聚苯乙烯具塞离心管中,加入1%醋酸15 mL,涡旋使其充分浸润,放置30 min,精密加入15 mL乙腈与100 μL内标溶液,涡旋混匀,剧烈振荡(500次/min) 5 min,加入无水硫酸镁与无水醋酸钠(质量比4∶1)的混合粉末7.5 g,立即摇散,再次剧烈振荡(500次/min)3 min,于冰浴中冷却10 min,离心(4 000 r/min)5 min,待净化。精密移取上清液9 mL置于分散固相萃取净化管(预装300 mg PSA、300 mg C18、90 mg GCB、600 mg Silica、900 mg无水硫酸镁)中,涡旋混匀,剧烈振荡(500次/min) 5 min,离心(4 000 r/min)5 min,精密移取上清液5 mL,置氮吹仪上于40 ℃水浴浓缩至约0.4 mL,加乙腈定容至1 mL,涡旋混匀,过0.22 μm微孔滤膜,取续滤液,即得。

2 结果与讨论

2.1 前处理方法的优化

早期QuEChERS法[8]采用乙腈提取,氯化钠盐析,该方法对大多数农药的回收率良好,但对部分碱性条件下不稳定农药(如百菌清和p,p'-滴滴涕等)的测定结果重现性差、加标回收率低,且部分农药在标准溶液中不稳定。针对上述问题,AOAC 2007.01方法[18]改用pH 5.0的醋酸盐缓冲体系,用含少量醋酸的乙腈配制标准溶液。本文在中国药典2015版四部通则<2341>方法[16]的基础上对样品前处理方法进行优化。

2.1.1 药材浸泡时间的选择三七的含水量低,且部分农药在碱性或中性条件下易降解,因此采用1%醋酸浸泡三七药材。以检出残留量较大的嘧菌酯、苯醚甲环唑、戊唑醇、氟环唑与嘧霉胺的三七阳性样品为研究对象,考察了不同浸泡时间(0、0.5、1、2 h)对提取效率的影响。结果表明,随着浸泡时间的增加,各农药的提取效率呈增加趋势,但浸泡0.5 h后提取效率相差不大,说明浸泡0.5 h即可将农药提取完全,因此选择浸泡时间为0.5 h,与中国药典2015版四部通则<2341>方法一致。

2.1.2 净化材料的选择在中国药典2015版四部通则方法的净化材料(即300 mg PSA、300 mg C18、90 mg GCB、300 mg Silica)基础上,进一步优化了净化材料配比。以三七提取液中所含的主要皂苷类成分为指标,采用中国药典2015版的高效液相色谱法测定净化前、后提取液中三七皂苷R1、人参皂苷Rg1、人参皂苷Rb1、人参皂苷Re总量,评价净化效果。图1结果显示,GCB用量对皂苷总量几乎无影响,C18对皂苷的去除效果不明显,但两者对三七中其他干扰成分有一定净化作用,故选择上述药典方法的用量。Silica与PSA对总皂苷的去除效果最为明显,但当PSA用量增至300 mg以上时,皂苷总量下降不明显;随着Silica用量的增加,皂苷总量下降明显,而Silica用量超过600 mg时,净化材料所占体积增加,无法从净化管中移取5 mL净化液进行浓缩,故PSA和Silica用量分别选择为300、600 mg。

综上,最终确定净化材料的配比为300 mg PSA、300 mg C18、90 mg GCB、600 mg Silica。此时,净化步骤总皂苷的去除率达94.3%,净化效果较好。优化条件下508种农药的总离子流色谱图见图2。

2.2 基质效应的评价

基质效应是痕量分析的常见现象。样品基质中的共提取干扰物影响待测化合物的仪器响应值,使得相同浓度的目标化合物在溶剂标准溶液与基质标准溶液的响应值不一致,表现为基质标准溶液的仪器响应发生增强或抑制。本文采用下式对目标化合物的基质效应进行评价:基质效应=[(基质匹配标准曲线的斜率/溶剂标准曲线的斜率)-1]×100%[19]。结果发现,仅有136种农药的基质效应不明显(<20%),372种农药均产生中等或较强的基质抑制效应(≥20%)。为克服基质效应的影响,采用基质匹配内标标准曲线法进行定量。

2.3 方法学验证

2.3.1 线性关系与检出限按照“1.2”方法配制基质混合标准工作溶液,采用本方法进行前处理和测定。以各组分的质量浓度为横坐标,对应峰面积与内标峰面积的比值为纵坐标,进行回归分析。结果表明,508种农药均在0.005~0.4 mg/L范围内呈良好线性关系,相关系数(r)均大于0.99。由于待测农药数量多,难以逐一稀释至每个农药的最低检出浓度,因此以实际检测到各农药2个MRM反应的标准曲线最低浓度作为检出限,得到方法检出限(LOD)为0.005~0.1 mg/kg(见表1)。

2.3.2 回收率与相对标准偏差将混合标准储备溶液添加到三七空白样品中,按“1.4”方法分别制得加标水平为10、50、100、200 μg/kg的样品溶液,每个水平重复7次。其中,100 μg/kg加标水平下的平均回收率和相对标准偏差(RSD)见表1,除8种农药外,其余500种农药的回收率为50.2%~117%。

对各加标水平下508种农药的回收率百分比进行分析。结果显示,10 μg/kg加标水平下有412种农药的回收率为70%~120%,占农药总数的81%;50 μg/kg加标水平下有485种农药的回收率为70%~120%,占总数的95%;100 μg/kg加标水平下有468种农药的回收率为70%~120%,占总数的92%;200 μg/kg加标水平下有477种农药的回收率为70%~120%,占总数的94%。各加标水平下的RSD均不大于15%。

2.4 实际样品分析

3 结 论

本文采用改良的QuEChERS方法对样品进行提取与净化,建立了快速检测三七中508种农药残留的液相色谱-串联质谱方法。本方法样品前处理简便、快速,灵敏度与准确度满足农药残留检测要求,可用于三七中农药多残留的快速测定,为三七中农药的合理规范使用提供了技术参考。
